制备方法


2-(5-乙基-2-吡啶基)乙醇(Ⅰ)和对氟硝基苯缩合,得到化合物(Ⅱ)。接着还原为胺(Ⅲ),然后在含氢溴酸的丙酮-甲醇中重氮化,再加入丙烯酸甲酯偶合得到化合物(Ⅳ)。继而和硫脲缩合环合,再水解得到吡格列酮。

2.胰岛素增敏剂,为新一代Ⅱ型糖尿病治疗药,副作用小。可减少外周组织和肝脏的胰岛素抵抗,增加依赖胰岛素的葡萄糖的处理,并减少肝糖的输出。吡格列酮可改善胰岛素抵抗患者的胰岛素敏感性,提高胰岛素对细胞的反应性,并改善体内葡萄糖平衡障碍。用于治疗糖尿病。
