制备方法


1、二环己基硫脲法工业上由环己胺与二硫化碳反应生成N,N'-二环己基硫脲,经脱硫化氢而得。 2、二环己胺尿素法也可由N,N'-二环己基脲与五氧化二磷在吡啶中反应而得。 4、制N,N’一二环已基脉 将240克(2.42摩尔)环已胺与60克(1 .00摩尔)尿素在480毫升异戊醇中回流反应20小时。冷后滤出晶体,乙醚洗涤,于燥,得N,N’一二环已基豚200克,产率89%。 5、制二环已基碳二亚胺 将200克(0.891摩尔)二环已基脉,300克(1 .574摩尔)一甲苯磺既氯在600毫升无水吡啶中于70℃搅拌反应l小时。将反应物倒入1.5公斤碎冰中,用乙醚萃取。乙醚液用水洗涤、干燥。蒸去乙醚,残余物经减压蒸锚。得二环已基碳二亚胺152克(82%)。

用作生化试剂和有机合成脱水缩合剂。

用途


1、用于阿米卡星及氨基酸的合成脱水,是一种很好的低温生化脱水剂,也用于酸、酐、醛、酮等的合成。在日本,用于谷胱甘肽的脱水剂,占总消费的90%。该品作为脱水缩合剂时,可在常温下经短时间反应即成,反应后产物为二环己基脲。由于该产物在有机溶剂中溶解度很小,所以反应产物易于分离;同时,由于该品很难溶于水,因此即使是在水溶液中,反应仍然可以进行下去。该品还用于肽、核酸的合成,使用本品可以在室温下很容易由具有游离羧基的化合物和具有游离氨基的化合物合成肽,并且收率很高。用于生产增血压素、环磷酸腺苷。

2、1,3-二环己基碳二亚胺(DCC)是一种很强的脱水试剂,多用于酰胺、酯和酸酐的合成中。同时,DCC在醇氧化为酮、一级酰胺转化为腈、β-羟基酮转化为α,β-不饱和酮的反应中得到了比较广泛的应用。

酰胺、酯、酸酐等的合成 (脱水剂)  DCC常用在多肽合成以及一些胺类与羧酸反应形成酰胺键的化合物中,其反应产率较高,反应速率也较快 (式1)[1]

在酯的合成中,伯醇、仲醇、烷基硫醇等均可在DCC存在的情况下与羧酸类化合物发生偶合反应。而对空间位阻较大的醇类来说,反应的产率相对较低 (式2)[2]。磷酸酯[3]也可在DCC的偶合下进行合成。

对于酸酐的合成,DCC是一种最简便、反应条件最温和,同时产率最高的偶合试剂[4]

醇到醛或酮的转化  在DCC存在下,DMSO可被活化,进而与伯醇或仲醇反应生成相应的醛或酮,从而实现由醇到醛或酮的转化 (式3),与其它金属催化的氧化反应相比,DCC存在下的氧化反应条件极其温和,产物单一,同时具有较好的化学选择性[5]

脱水反应  DCC具有很强的脱水性,因此也常被用来合成具有某些特殊官能团的化合物,如使用羧酸合成烯酮的反应 (式4)[6]β-羟基酮、β-羟基酯等与DCC反应可以生成α,β-不饱和酮或酯。

硫脲转化为氰基胍  在DCC的作用下,硫脲可以在适当的反应条件下转化为氰基胍 (式5)[7]

羧酸与NHS的反应形成活化酯  在DCC的偶合下,羧酸与NHS反应形成活化酯 (式6)[8],然后氨基或羟基对羰基的亲核进攻形成酰胺键或酯键。
